    CUSABIO transfected the expression vector which inserted the recombinant DNA into the yeast, cultured the cells, and then induced the transcription and translation of the cloned vector. The N-terminal 6xHis tag sequence was appended to the gene coding for the yeast of the Pig C5 protein to form the recombinant DNA. The recombinant Pig C5 was expressed as N-terminal 6xHis-tagged fusion. The purity of the protein is greater than 90% assayed by SDS-PAGE. It has an apparent molecular weight of approximately 11 kDa.

    C5a, a peptide derived from the fifth component of complement, caused significant prolonged smooth muscle contraction in isolated guinea pig trachea. Earlier studies highlighted an amplifying effect of locally applied C5a on the process; however, the role of systemic complement (C) activation with C5a liberation in blood has not been explored to date. C5a was shown to intensify the allergen-induced anaphylactic crisis in isolated, perfused guinea pig hearts. Inhibitors of C activation or C5a or adenosine action might be useful in preventing or ameliorating C activation-related cardiac anaphylaxis. Many findings unrevealed the effect of C5a anaphylatoxins on guinea-pigisolated blood vessels.

    Derived from proteolytic degradation of complement C5, C5a anaphylatoxin is a mediator of local inflammatory process. Binding to the receptor C5AR1 induces a variety of responses including intracellular calcium release, contraction of smooth muscle, increased vascular permeability, and histamine release from mast cells and basophilic leukocytes. C5a is also a potent chemokine which stimulates the locomotion of polymorphonuclear leukocytes and directs their migration toward sites of inflammation.
