H3L
H3L 蛋白全稱為組蛋白 H3 樣蛋白,也常被稱作 H3 樣變體、H3.3 樣組蛋白,是染色質核小體的重要組成部分,參與 DNA 折疊、基因表達調控與細胞分化維持,在胚胎發育與神經細胞成熟中尤為關鍵。它的結構與功能異常會引發染色質穩態失衡,與兒童彌漫中線膠質瘤、神經發育障礙、Bryant-Li-Bhoj 綜合征等疾病密切相關,也在多種實體瘤的表觀遺傳紊亂中扮演重要角色。目前,針對 H3L 相關異常的靶向藥物以表觀遺傳調節劑為主,甲基轉移酶抑制劑、組蛋白去乙酰化酶抑制劑等已進入臨床研究,同時靶向 H3 突變體的免疫治療與小分子精準藥物也在快速推進,為相關難治性疾病提供新的干預方向。
